This postdoctoral position is part of a research project funded by a Wallenberg Scholar grant. The postdoctoral researcher will work in an interdisciplinary team with expertise in molecular biology, biochemistry and structural biology. The project aims to understand the molecular mechanisms that govern mitochondrial DNA replication and gene expression, and how defects in these processes contribute to human disease. Subject area Structural studies of mitochondrial DNA replication Subject area description Mitochondrial DNA replication is essential for maintaining mtDNA integrity and stability and thereby for normal mitochondrial function and cellular energy production. Defects in the replication process can lead to mtDNA depletion or deletions and cause mitochondrial disease. Our research aims to understand the molecular mechanisms that govern replication of human mtDNA. We combine structural biology, in vitro biochemistry and cell biology to investigate how the mitochondrial DNA replication machinery is organized and regulated. A particular focus is on structural and functional studies of the protein complexes involved in mtDNA replication and on understanding how disease-associated mutations affect these processes. Job assignments In this project, In this project, the postdoctoral researcher will carry out structural and functional studies of protein and protein-DNA complexes involved in mitochondrial DNA replication. A central aim will be to determine structures of replication complexes using single-particle cryo-electron microscopy (cryo-EM) and use these structures to understand how the proteins interact and regulate mtDNA replication at the molecular level. The work will include preparation and biochemical characterization of protein complexes, cryo-EM sample preparation and grid optimization, as well as processing and structural analysis of cryo-EM data. Structural findings will be combined with in vitro biochemical analyses to test mechanistic models and investigate how disease-associated mutations affect the function of the replication machinery. The postdoctoral researcher is expected to drive the project independently while working closely with other members of the research group and our collaborators. Through large-scale whole-genome sequencing and other high-throughput approaches, these processes can be mapped in detail, providing insights into tumor initiation, progression, and treatment response. This position focuses on the application and development of bioinformatics methods for the analysis of genomic cancer data, including large public international datasets and locally generated patient-derived sequencing data. Analysis of p38 MAP kinase inhibitors to improve cisplatin function : The Peter Naredi lab studies the action of the anti-cancer drug cisplatin with focus on the cell biology basis for its function and to identify signalling pathways that modulate its response in the nematode C.elegans. Cisplatin is a commonly used anti-cancer drug whose cytotoxic effects are regulated by intracellular signaling pathways. One of the pathways that has been identified is the p38 MAP Kinase pathway. Knockdown of the pathway leads to enhanced cisplatin sensitivity The p38/MAP kinase pathway has been identified as an important regulator of cisplatin response in both C. elegans and human tumor cells. Understanding how this pathway can be modulated may improve treatment strategies. Using a humanized p38MAP Kinase model, the lab has tested inhibitors with the aim to conduct translational research into making cisplatin more effective. Similar work is also conducted on another genetic pathway that modulates cisplatin response. Duties The teaching assistent will assist with ongoing research projects in the laboratory and contribute to experimental work. Duties will include: • Performing drug treatment experiments and cytotoxicity assays. • Conducting gene expression analyses using quantitative PCR (qPCR). • Performing protein analyses, including Western blotting. • Assisting with fluorescence microscopy experiments and image acquisition. • Maintaining accurate and up-to-date laboratory records and documentation. • Presenting experimental results and progress updates at research group meetings. We are currently seeking a candidate for a postdoctoral position in colorectal cancer (CRC) research. The project will explore the role of goblet cell-intrinsic mucosal defence systems in determining susceptibility to CRC. The candidate will join a newly established Wallenberg Centre for Molecular and Translational Medicine (WCMTM) at the University of Gothenburg (www.wcmtm.gu.se). As part of this venture, Dr. George Birchenough has established his research group (www.birchenoughlab.org) at the Department of Medical Biochemistry & Cell Biology at the University of Gothenburg. The group works towards a better understanding of how mucosal defence systems control our interactions with the environment, how those systems fail in disease and how they might be prophylactically or therapeutically targeted. The group is part of a large network of researchers at Gothenburg University, most importantly the Mucin Biology Groups cluster (www.medkem.gu.se/mucinbiology) that currently includes over 30 researchers working at the forefront of mucosal biology. The candidate will have the opportunity to collaborate with experts in multiple experimental methods that work within the cluster, as well as more widely with other local and national groups within the WCMTM network. Subject area Translational Mucosal Biology Subject area description Our mucosal surfaces are the interface between the environment and our tissues. Their physiological functions are critical, but necessitate constant exposure of living cells to a vast collection of microbial lifeforms that includes many opportunistic and obligate pathogens. As a result, our mucosa has evolved multiple defensive systems that allow them to endure in this environment, and failure of these systems is often linked to development of acute and chronic disease. Consequently, understanding the development and regulation of these systems is key to understanding the pathoaetiology of mucosal disease and how they can be prevented or treated. Colorectal cancer (CRC) is one of the most frequent and lethal types of cancer, and is especially prevalent in nations with a high human development index, indicating that factors associated with a modern, Westernized lifestyle are linked to CRC development. Most cases of CRC result from spontaneous mutations rather than heritable oncogenic mutations, and dysregulated interactions between the intestinal microbiota and immune system are implicated in tumorigenesis. Goblet cell-intrinsic defence systems, such as the mucus layer and sentinel goblet cells normally control host-microbiota interactions, but we have recently found that these systems are highly susceptible to environmental alterations caused by exposure to a Western-style diet. Thus, disruption of goblet cell-dependent functions may have a causal relationship with CRC development. By combining data from pre-clinical mouse models with analysis of samples from healthy and CRC patients, we hope to develop a better understanding of how goblet cell functions contribute to CRC susceptibility and develop new approaches to prevent CRC development. Duties The successful candidate will be expected to work extensively with pre-clinical mouse models and human tissues acquired during routine surgery. The candidate will be encouraged and expected to conduct their experimental research in an independent manner and to make significant contributions to project planning. The candidate will also actively contribute to the local research environment at the Mucin Biology Groups and engage in interactions with collaborators and other groups belonging to the WCMTM network. If necessary, the candidate may also be asked to assist in the supervision of Masters or PhD students. Subject area Chemistry with a specializing in chemical glycobiology Subject area description Chemical glycobiology concerns the development and application of chemical approaches to understand the structures, dynamics and biological functions of glycans and glycoconjugates. The field integrates synthetic and analytical chemistry with chemical biology and molecular and cell biology to investigate how glycosylation influences molecular recognition, signalling, membrane organisation and biological function in health and disease. Relevant approaches include carbohydrate and bioconjugation chemistry, metabolic and bioorthogonal labelling, chemical probe design, glycan editing, and mass-spectrometry-based glycomics and glycoproteomics. By enabling the identification, characterisation and manipulation of glycans and glycoconjugates in complex biological systems, chemical glycobiology provides a molecular link between chemistry, glycobiology and membrane biology. At the Department of Chemistry and Molecular Biology, the subject will complement and connect existing strengths in medicinal and analytical chemistry, biochemistry, glycobiology, proteomics and membrane biology. Available department infrastructure includes advanced microscopes, facilities for culturing mammalian cells and microbes, high throughput/high content phenotypic cell-based screening, equipment for purification of small molecules and macromolecules, NMR, lasers, electron microscopy, and imaging mass spectrometry. An important opportunity is to integrate experimental glycoscience with computational and AI-based approaches for predicting and interpreting molecular structure, interactions and function, in interaction with existing research groups in the department. Project description: Ulcerative colitis is a chronic inflammatory bowel disease characterised by periods of remission and relapse. Although modern treatment often leads to clinical, endoscopic and histological healing, many patients relapse. Why an apparently healed intestinal mucosa remains biologically vulnerable is a central unanswered question. This PhD project aims to identify the molecular mechanisms that determine whether remission remains stable or the disease relapses. The project is based on the hypothesis that the intestinal microenvironment, shaped by interactions between the microbiota, microbial metabolites and host regulatory signals, is not fully restored during remission, which may increase susceptibility to renewed inflammatory stimuli. The PhD student will be part of BioLabX, Sweden's national research school in Biomedical Laboratory Science. The project is particularly relevant for applicants with a background in Biomedical Laboratory Science. The PhD student will combine patient-oriented research with experimental models to investigate how the intestinal microenvironment influences immune function during remission. The work includes handling and analysis of human patient samples, functional cell models using human macrophages, advanced cell culture, flow cytometry and molecular biology, as well as metabolomics and lipidomics. Bioinformatic and statistical methods will be used to integrate molecular data with clinical follow-up and identify biomarkers that may predict relapse. The research environment is translational and interdisciplinary, with close collaboration between preclinical researchers, clinical gastroenterologists and national research networks. Through BioLabX, the PhD student will have access to a national network, specialised doctoral courses and pedagogical training. The PhD student will also participate in teaching Biomedical Laboratory Science in Swedish. Eligibility Doctoral education requires general eligibility and, where appropriate, specific eligibility as set out in the general syllabus for the subject. Admission and employment Once you have been admitted to doctoral studies, you will be employed as a doctoral student at the University of Gothenburg. The provisions for employment as a doctoral student can be found in ordinance SFS 1993:100. Initial employment as a doctoral student may apply for a maximum of one year, and may be renewed by a maximum of two years at a time. A doctoral student may be employed as a doctoral student for a maximum of eight years, but the total period of employment may not be longer than the equivalent of full-time education at doctoral level for four years. The group of Professor Marc Pilon performs basic research on molecular pathways that regulate lipid metabolism and membrane homeostasis using mostly the nematode C. elegans as model organism. Duties The postdoctoral fellow would be expected to carry out independent/semi-independent research leveraging the collection of C. elegans mutant alleles and/or the expertise of the Pilon group to further advance our understanding of lipid/membrane homeostasis. Task will likely include hypothesis formulation, setting and performing forward genetics screens using C. elegans and characterization of pathways using combinations of genetic interactions studies, microscopy, CRISPR/Cas9 genome editing among other methods. Qualifications To be eligible for appointment as a postdoctoral researcher, an applicant must have obtained a doctoral degree, or a foreign qualification deemed equivalent to a doctoral degree.
